My project is focused on exploring the immune signatures that characterize juvenile systemic sclerosis (jSSc) that may predict clinical response, non-response, or disease flare after autologous stem cell transplantation (ASCT) and CD19-directed CAR-T cell therapy (CART19). To determine potential biomarker signatures, we will start by better defining the extended T cell and B cell immunophenotype of jSSc at baseline utilizing PBMCs collected pre and post stem cell transplant at specific time points starting with multiparameter Flow cytometry. Later will attempt to characterize BCR and TCR repertoire remodeling after ASCT and CART19 though high throughout sequencing.
